The expression level of each gene must be precisely controlled to ensure proper cell fate and function. At the same time, gene expression needs to be very flexible to allow rapid changes in protein levels for cells to adapt to their continuously changing environment. We have developed live-cell imaging technology to visualize individual steps in gene expression in real-time at the single molecule level, which we combine with new types of genetic engineering to study molecular mechanisms that underlie precise gene expression control, and we aim to understand how control of gene expression affects cell fate decisions. We have also developed single-molecule imaging approaches to visualize viral gene expression and are applying these technologies to study viral infection, translation and replication in real-time.
